هدف: این پژوهش با هدف ارزیابی اثر پرایمینگ بذر با محرک‌های غیرزیستی شامل اسید سالیسیلیک، پرولین، گابا (γ-آمینوبوتیریک اسید) و ملاتونین بر بهبود تحمل تنش شوری در گل همیشه بهار انجام شد.
مواد و روش‌ها: آزمایش به‌صورت فاکتوریل در قالب طرح کاملاً تصادفی تحت شرایط کنترل‌شده آزمایشگاهی با سه تکرار اجرا شد. تیمارها شامل چهار سطح شوری (0، 3، 6 و 9 دسی‌زیمنس بر متر) و شش نوع پرایمینگ بذر (شاهد بدون پرایمینگ، هیدروپرایمینگ با آب مقطر، اسید سالیسیلیک 5/1 میلی‌مولار، پرولین 100 میلی‌گرم در لیتر، گابا 10 میلی‌مولار و ملاتونین 100 میکرومولار) بودند. بذرها به مدت 24 ساعت در تاریکی و دمای 20 درجه سلسیوس پرایم شدند و سپس تحت تنش شوری قرار گرفتند. صفات مورد بررسی شامل درصد و سرعت جوانه‌زنی، شاخص بنیه گیاهچه، طول ریشه‌چه و ساقه‌چه، و وزن خشک اندام هوایی و ریشه بودند.
یافته‌ها: افزایش سطح شوری موجب کاهش معنی‌دار تمام صفات جوانه‌زنی و رشد گیاهچه شد، به‌طوری‌که درصد جوانه‌زنی در سطوح شوری 3، 6 و 9 دسی‌زیمنس بر متر به‌ترتیب 7/6، 9/18 و 5/24 درصد نسبت به شاهد کاهش یافت. پرایمینگ بذر به‌طور معنی‌داری اثرات منفی شوری را کاهش داد. در میان تیمارها، هیدروپرایمینگ به‌طور پیوسته درصد و سرعت جوانه‌زنی، شاخص بنیه گیاهچه و تجمع زیست‌توده را در همه سطوح شوری بهبود بخشید و حتی در شوری شدید (9 دسی‌زیمنس بر متر) نیز اثر مثبت خود را حفظ کرد. گابا و اسید سالیسیلیک بیشترین افزایش وزن خشک اندام هوایی را ایجاد کردند، در حالی که پرولین، ملاتونین و گابا نیز رشد اولیه گیاهچه را بهبود بخشیدند. در شرایط شوری شدید، گیاهان پرایم‌شده کاهش عملکرد کمتری با افزایش سطح نمک نشان دادند که بیانگر بهبود تحمل به تنش است.
نتیجه‌گیری: به‌طور کلی، پرایمینگ بذر با محرک‌های غیرزیستی مناسب، راهکاری مؤثر، اقتصادی و سازگار با محیط‌زیست برای کاهش خسارات ناشی از شوری و بهبود استقرار و بهره‌وری گیاهان دارویی در محیط‌های شور است. اثر مثبت تیمارهای پرایمینگ به نوع محرک و شدت تنش شوری وابسته بود.
جنبه‌های نوآوری:
  • پرولین و گابا اثرات بازدارنده شوری بر جوانه‌زنی همیشه بهار را کاهش دادند.
  • هیدروپرایمینگ تجمع زیست‌توده را در همه سطوح شوری بهبود بخشید.
  • اثربخشی محرک‌ها به شدت شوری و نوع صفت وابسته بود.
