چکیده مبسوط
مقدمه: یکی از دلایل عمده کاهش محصول در گیاهان زراعی هجوم علف‌های هرز است. دگرآسیبی یکی از توانایی‌های علف‌های هرز است که معمولاً با اثرات بازدارندگی، بر رفتار جوامع گیاهی تأثیر می‌گذارد؛ بنابراین بررسی تأثیر دگرآسیبی بر گیاهان زراعی از اهمیت خاصی برخوردار است. همچنین استفاده از گیاهان دگرآسیب و شناسایی ترکیبات زیست‌فعال آن‌ها می‌تواند راه‌کارهای مناسبی برای مدیریت علف‌های هرز نشان دهد؛ بنابراین در این تحقیق ضمن بررسی تأثیر مواد دگرآسیب موجود در ریشه گیاه آلاله داسی روی شاخص‌های جوانه‌زنی بذرگندم، نحوه عمل آن بر برخی از فعالیت‌های آنزیمی مطالعه و ترکیبات احتمالی شرکت‌کننده در دگرآسیبی این گیاه، شناسایی و گزارش شده است.
مواد و روش‌ها: آزمایش‌های جوانه‌زنی و رشد گیاهچه بذر گندم رقم سرداری تحت تأثیر تیمار عصاره آبی ریشه آلاله داسی در پنج سطح، شاهد (آب مقطر)، 5، 10، 15 و 20 درصد عصاره به­صورت طرح کاملاً تصادفی با چهار تکرار در آزمایشگاه پژوهشی تولید و ژنتیک گیاهی دانشگاه مراغه طی سال 1397 انجام شد. هم‌زمان با بررسی اثر تیمار بر مجموعه صفات جوانه‌زنی و گیاهچه، تغییر در فعالیت آنزیم‌های آلفا آمیلاز، کاتالاز، پراکسیداز، سوپر اکسید دیسموتاز و فنل اکسیداز نیز در این مرحله بررسی شدند. همچنین، شناسایی ترکیبات شیمیایی عصاره متانولی ریشه توسط GC/MS انجام شد.
یافته‌ها: با توجه به نتایج، با افزایش غلظت عصاره ریشه آلاله داسی درصد و سرعت جوانه‌زنی بذر گندم به‌طور معنی‌دار کاهش یافت و در غلظت بیش از 15 درصد جوانه‌زنی متوقف شد. نتایج رشد گیاهچه حاکی از بازدارندگی شدید عصاره ریشه بر رشد اندام‌های ریشه‌چه و ساقه‌چه گندم بود که با کاهش معنی‌دار در طول و وزن این اندام‌ها همراه شد که درنهایت قدرت گیاهچه را کاهش داد. همچنین عصاره ریشه، فعالیت آنزیم آلفا آمیلاز را به‌طور معنی‌دار کاهش داد ولی فعالیت آنزیم‌های آنتی‌اکسیدانت در سطوح پایین عصاره افزایش و در سطوح بالاتر کاهش یافتند. در تجزیه شیمیایی، ترکیبات Octadecatrienal، Dihydro-4H-pyrazolo[3,4-d]pyrimidin-4-one، Hexadecanoic acid، Hexadecanoic acid, 2-hydroxy-1(hydroxymethyl)ethyl ester، Isopropyl isothiocyanate و Cyclohexanone با مقادیر بالا شناسایی شدند که بازدارنده جوانه‌زنی نیز هستند.
نتیجه‌گیری: عصاره ریشه آلاله داسی اثر بازدارندگی بسیار قوی روی قوه و قدرت نامیه بذر گندم داشت. بر اساس یافته‌های پژوهش نحوه عمل ترکیبات دگرآسیب آلاله داسی از طریق القاء تنش اکسیداتیو و ممانعت از تحریک مواد اندوخته بذری در هنگام جوانه‌زنی می‌باشد. حساسیت بالای فعالیت آنزیم آلفاآمیلاز به مواد دگرآسیب در این آزمایش ردیابی شد. همچنین کاهش فعالیت تمامی آنزیم‌های مورد مطالعه در غلظت‌های بالا قابل توجه بود. مشتقات مربوط به اسید استئاریک و اسید پالمتیک در حدود 30 درصد ترکیبات را به خود اختصاص دادند که با احتمال بسیار قوی تداخل آنها با فعالیت آنزیم‌ها وجود دارد.

جنبه‌های نوآوری:
1- تأثیر دگرآسیبی عصاره آبی ریشه آلاله داسی بر روی شاخصه‌های جوانه‌زنی گندم زراعی بررسی شد.
2- شناسایی ترکیبات موجود در عصاره متانولی ریشه آلاله داسی صورت گرفت.
3- القاء تنش اکسیداتیو توسط ترکیبات دگرآسیب مشاهده شد.
