چکیده مبسوط                                                                                                                      
مقدمه: به‌منظور استفاده بیشتر از فصل رشد بهاری سازوکار‎های بهبود دهنده جوانه‏زنی گیاهان بهاره در دمای پایین­تر از دمای مناسب اهمیت زیادی دارد. از آنجا که یکی از راه‌های کاهش خسارت دمای پایین ارتقا سیستم آنتی‌اکسیدانی گیاهچه است، لذا در این آزمایش اثر اسید سالیسیلیک و جیبرلین بر جوانهزنی و سیستم آنتی‌اکسیدانی بذر گیاه کدوی پوست‌کاغذی تحت دماهای پایین مورد بررسی قرار گرفت.
مواد و روشها: آزمایش به‌صورت فاکتوریل شامل چهار غلظت جیبرلین (صفر، 250، 350 و 450 میلی‏گرم در لیتر) و چهار غلظت اسید سالیسیلیک (صفر، 5/0، 1 و 5/1 میلی­مولار) و سه سطح دما (8، 11 و 14 درجه سانتی­گراد) در قالب طرح کاملاً تصادفی در شرایط کنترل شده و در شش تکرار در دانشگاه شهرکرد در سال 1396 اجرا شد. بذرها پس از ضدعفونی در ظروف حاوی محلول‌های با غلظت صفر، 250، 350 و 450 میلی‏گرم در لیتر جیبرلین و محلول‌های با غلظت صفر، 5/0، 1 و 5/1 میلی­مولار اسید سالیسیلیک قرار گرفتند و به مدت 24 ساعت در شرایط تاریکی و در دمای 15 درجه ‌سانتی‌گراد در اتاقک رشد قرار داده شدند. سپس شستشوی بذرها انجام شده و در دماهای مورد نظر قرار داده شدند، جوانه­زنی هر 24 ساعت بر مبنای خروج ریشه­چه به اندازه 2 میلی­متر ثبت گردید. در پایان روز هشتم پس از جداسازی گیاهچه­های طبیعی و غیرطبیعی تعداد 20 گیاهچه طبیعی از هر پتری انتخاب و سپس پارامترهای سرعت و درصد جوانه­زنی، پروتئین محلول، میزان مالون دی آلدئید، فعالیت آنزیم سوپر اکسید دیسموتاز، آنزیم گایاکول پرکسیداز و آنزیم کاتالاز اندازه‎گیری شد. برای مقایسه میانگین‎‎ها از آزمون LSD در سطح احتمال پنج درصد استفاده شد.
یافته‌ها: نتایج نشان داد در دمای 8 درجه سانتی­گراد هیچ یک از تیمارهای به کار برده شده نتوانست به جوانه­زنی گیاه کمک کند و به همین علت از آزمایش حذف شدند، در دمای 11 درجه سانتی­گراد استفاده از تیمار سالیسلیک اسید یک میلی­مولار و در دمای 14 درجه سانتی­گراد 350 میلی‏گرم در لیتر جیبرلین بیشترین افزایش سرعت و درصد جوانهزنی را نسبت به شاهد نشان داد. در دمای 11 درجه سانتی­گراد فعالیت آنزیم­های آنتی‌اکسیدانی بیشتر تحت تأثیر هورمون جیبرلین قرار گرفت به­طوری که بیشترین میزان فعالیت آنزیم سوپراکسیددیسموتاز در تیمار 350 میلی‏گرم در لیتر و بیشترین میزان فعالیت آنزیم­های کاتالاز و گایاکول پراکسیداز و کمترین میزان پروتئین محلول در تیمار 250 میلی‏گرم در لیتر جیبرلین مشاهده شد. در دمای 14 درجه سانتی­گراد هورمون اسید سالیسیلیک موفق­تر بود و اسید سالیسیلیک 5/1 میلی­مولار میزان فعالیت آنزیم سوپراکسیددیسموتاز، اسید سالیسیلیک 5/0 میلی­مولار میزان فعالیت آنزیم کاتالاز و اسید سالیسیلیک 1 میلی­مولار میزان فعالیت آنزیم گایاکول پراکسیداز را افزایش داد، همچنین این هورمون در راستای کاهش میزان پروتئین محلول نیز موفق بود.
نتیجه‎گیری: در این آزمایش تحمل گیاهچه به دماهای پایین توسط پیش‌تیمارهای جیبرلین و اسید سالیسیلیک به اثبات رسید. به‌طورکلی نتیجه‎گیری می‎شود که کاربرد جیبرلین و اسید سالیسیلیک باعث افزایش فعالیت آنزیمهای آنتی‌اکسیدان و درنتیجه تحمل گیاهچه کدوی پوست کاغذی به تنش دمای پایین می­شود و بنابراین با استفاده از این ماده میتوان اثر سرماهای احتمالی در ابتدای فصل رشد این محصول را تعدیل نمود.

جنبه‌های نوآوری:
1- گیاهچه کدوی پوست کاغذی توسط پیش تیمارهای جیبرلین و اسید سالیسیلیک به دماهای پایین متحمل می‎شود.
2- کاربرد جیبرلین و اسید سالیسیلیک باعث افزایش فعالیت آنزیمهای آنتی‌اکسیدان می­شود.
3- با استفاده از جیبرلین و اسید سالیسیلیک میتوان اثر سرماهای احتمالی در ابتدای فصل رشد این محصول را تعدیل نمود.
