چکیده مبسوط
مقدمه: استویا گیاهی چندساله، روز کوتاه و متعلق به خانواده آستراسه است که گیاه شکر برگ نیز نامیده می­‌شود. جوانه‌­زنی ضعیف در این گیاه مانعی برای کشت در مقیاس بزرگ بوده و سبب کمیاب شدن و گران‌قیمت بودن مواد مؤثره این گیاه دارویی شده است. در بسیاری از گیاهان مرحله جوانه­‌زنی بذر به شوری حساس بوده و تعیین کننده بقای گیاهان در خاک‌های شور می­ باشد. سطوح بالای شوری خاک می­‌تواند به طور معنی داری از جوانه‌­زنی و رشد گیاهچه به دلیل اثرات مربوط به پتانسیل اسمزی بالا و سمیّت ویژه یون، جلوگیری نماید. از طرفی جاسمونات­‌ها از تنظیم کننده­‌های رشد گیاهی جدید به شمار می‌­روند که در افزایش مقاومت گیاهان به تنش­های محیطی از جمله تنش شوری نیز از نقش مهمی برخوردارند. به همین دلیل این آزمایش با هدف بررسی تأثیر پیش تیمار بذر با متیل جاسمونات بر شاخص­ های جوانه‌­زنی و صفات بیوشیمیایی گیاهچه­‌های گیاه دارویی استویا تحت تنش شوری انجام گرفت.
مواد و روش­ها: این تحقیق به صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار در سال 1395 در آزمایشگاه زیست‌­شناسی گیاهی مجتمع پروفسور حسابی، دانشگاه آزاد اسلامی واحد اسلام شهر اجرا شد. فاکتورهای آزمایش شامل پیش تیمار متیل جاسمونات در 5 سطح (صفر، 5/2، 5، 10 و 15 میکرومولار) و تنش شوری در 4 سطح (صفر، 3، 6 و 9 دسی زیمنس بر متر) فاکتورهای آزمایشی بودند. در پایان آزمایش صفات درصد جوانه‌­زنی و سرعت جوانه­‌زنی، میانگین زمان جوانه‌­زنی، ارزش جوانه‌زنی، طول گیاهچه، شاخص قوه نامیه بذر، کلروفیل کل، میزان پرولین، فعالیت آنزیم کاتالاز، پراکسیداز و سوپراکسیددیسموتاز اندازه­‌گیری شدند.
یافته­‌ها: نتایج تحقیق نشان داد که اثرات تنش شوری، متیل جاسمونات و اثر متقابل شوری و متیل جاسمونات بر درصد و سرعت جوانه­‌زنی، میانگین زمان جوانه‌­زنی، ارزش جوانه‌­زنی، شاخص بنیه بذر، کلروفیل کل، پرولین و فعالیت آنزیم کاتالاز معنی‌­دار بود. پرایمینگ بذر با 5 میکرومولار متیل جاسمونات در سطح شوری با هدایت الکتریکی صفر دسی زیمنس بر متر، بیشترین درصد و سرعت جوانه­زنی، ارزش جوانه‌­زنی، شاخص بنیه بذر، میزان کلروفیل کل و پرولین آزاد را داشت. با افزایش تنش شوری و میزان متیل جاسمونات فعالیت آنزیم کاتالاز افزایش معنی‌­داری نشان داد. شوری باعث کاهش شاخص‌­های جوانه‌­زنی و رشد گیاهچه استویا و افزایش فعالیت آنزیم­های پراکسیداز و سوپراکسیددیسموتاز گردید، ولی پرایمینگ بذر با متیل جاسمونات از طریق افزایش درصد و سرعت جوانه­‌زنی و شاخص بنیه بذر، رشد گیاهچه را بهبود بخشیده و اثرات ناشی از تنش شوری را تعدیل نمود.
نتیجه‌­گیری: به طور کلی با توجه به نتایج این تحقیق می­‌توان اظهار داشت که متیل جاسمونات به عنوان بهبود دهنده احتمالاً می‌­تواند باعث کاهش اثرات منفی شوری شود و با افزایش شاخص‌های جوانه­ زنی مثل درصد و سرعت جوانه‌­زنی در بهبود رشد گیاه استویا مؤثر باشد، البته برای دست‌یابی به نتایج دقیق­‌تر، باید تحقیقات بیشتری انجام گیرد.

جنبه­‌های نوآوری:
  1. تیمار شوری اثر منفی و متیل جاسمونات اثر مثبتی بر شاخص‌­های جوانه‌­زنی و فعالیت آنزیم­‌های آنتی‌اکسیدانت بذر استویا داشت.
  2. استفاده از متیل جاسمونات در غلظت 5 میکرومولار می‌­تواند در بهبود رشد گیاه استویا و کاهش اثرات منفی شوری مؤثر باشد.
