Abstract

In order to investigate the effects of priming with a KH2Po4 solution on emergence and heterotrophic growth of black cumin (Nigella sativa L.) seedling, a series of experiments were conducted at the Faculty of Agriculture, Ferdowsi University of Mashhad, Iran, in 2013 and 2014. A field experiment was conducted as factorial layout based on a randomized complete block design with four replications. The fertilizer resources (vermicompost (V)+Tiobacilus (T), micronized Sulfur (S)+T, V+S+T and control) and three levels of phosphorus (0, 30 and 60 kg.ha-1) were the first and second experimental factors, respectively. In the second experiment, selected treatments (in which showed emergence below 60% in the previous experiment) and seed priming (no priming, hydro priming and three levels of phosphorus (P)-priming as 100, 300 and 500 mM KH2Po4) were the first and second experimental treatments, respectively. Based on the results, the resources of soil amendment (V+T and V+S+T) significantly increased the emergence percentage of black seed (by 50 and 54%, respectively). In addition, seed reserve depletion percentage, seed reserve utilization efficiency and emergence percentage significantly increased by 300 mM KH2PO4 (by 9, 143 and 29%, respectively). There was a significant positive relationship between the weight of mobilized seed P and the emergence percentage of black seeds (R2= 0.90 **).

Abstract

Roselle is an important medicinal and industrial plant of the family of Malvaceae, and is planted in vast areas of Sistan and Baluchestan. In a laboratory study, the effect of varying temperatures on seed germination of Hibiscus sabdariffa was investigated and minimum, optimum and maximum temperatures for its germination were determined in a completely randomized design with four replications.  For this purpose, temperatures 5, 10, 15, 20, 25, 30, 35, 40, 45 and 50°C were considered in each treatment. Cardinal temperatures for germination were determined consistent with three models (i.e., Intersected-lines Model, Five-Parameters Beta Model and Quadratic Polynomial Model). The traits measured were germination percentage, the speed of germination and mean germination time. The temperature effect on all the measured traits was significant. The results of the regression analysis showed that the best model in terms of cardinal point of this plant is the Five-Parameters Beta Model. Given the results of this model, the minimum and the optimal temperatures for the germination of Roselle are 4.04°C, and 29.83° C, respectively.

Introduction:
 One of the biological methods of weed control is the use of allelopathic substances or herbal secretions that have the potential to inhibit growth weeds. Wheat allelopathic studies include the study of wheat allelopathy against other crops, weeds, pests and diseases, isolation and identification of allelopathic compounds, the effects of wheat toxicity on self-storage and management of wheat residues. Wild mustard (Sinapis arvensis) is a problematic weed in the country, especially in Golestan province. The present study was conducted to investigate the allelopathic potential of four wheat cultivars to use those cultivars in integrated weed management system to reduction growth mustard weed.
Materials and methods: A factorial experiment was conducted in completely randomized design with three replications at Physiology Laboratory of Golestan Agricultural Research Center. Treatments included concentrations of 0, 2.5, 5, and 7.5% aqueous extract of aerial and underground organs of four wheat cultivars (Morvarid, Moghan, Tajan, and Arta) with 6000 polyethylene glycol treatment in four concentration (zero, 2.5, 5, and 7.5 percent). Three-parameter logistic model was used to evaluate the allelopathic potential of wheat genotypes for reducing the percentage of wild mustard germination and liquid chromatography (HPLC) was used to determine the phenolic acids composition in wheat cultivar extract.
Results: Extract of shoot of all wheat cultivars reduced root length of wild mustard in comparison of control. Wheat cultivars reduced shoot length of wild mustard by 28% to 51% compared to control. The highest reduction in mustard shoot length was obtained from the Morvarid extract. Increase in shoot extract concentration of Arta to 7.5%, reduced root and shoot dry weight as 47 and 29% in compare of control treatment and Moghan cultivar in the same concentration (7.5%) caused reduction in root and shoot dry weight with 68 and 41% respectively. The highest reduction in germination indices of wild mustard was obtained from high concentrations of Morvarid and Moghan wheat extract. It was also found that with increasing concentration of polyethylene glycol (PEG) growth indices and germination components of wild mustard seed decreased, but this decrease was not significant. This confirms that the osmotic potential of the extract concentration is not involved in the exacerbation of the allelochemical effect and the likelihood of osmotic effect seems poor. In total, Morvarid cultivar with 25.34 mg phenolic acid content had the highest phenolic acid concentration, which was approximately three times more than that of Tajan, 1.5 times Moghan and three times of Arta cultivar. Morvarid cultivar had higher concentration of vanillic acid and ferulic acid than the other three cultivars. Overall, the least allelopathic effect was observed in Tajan cultivar and the most allelopathic effect was in Morvarid cultivar.
Conclusion: Among the cultivars studied, the highest inhibition was related to Morvarid cultivar. The results also showed that the highest amount of inhibition was related to shoot extract so that the root extract inhibition was less than the shoot on studied traits. In general, the results of the present study showed that some wheat cultivars have greater inhibitory potential against wild mustard weed, which can be used in breeding programs to produce cultivar with high allelopathic potential. Awareness of this issue is more important, especially in sustainable weed-management. Therefore, it is suitable to perform comprehensive studies on their allelopathic potential so that they can be used in agriculture, including combating with weeds, pests and plant diseases, breeding crop and horticulture, design herbicides and pesticides that is provide environmentally friendly, safe, and biodegradable.

Abstract

Introduction: Galbanum is one of the most important native medicinal plants in Iran, and nowadays the use of its products and derivatives has become widespread in various industries. Thus, the excessive harvest of this species within its natural habitats has increased. Therefore, the species is enlisted as prone to extinction. Due to the lack of information about germination behavior and dormancy alleviating or breaking methods of Galbanum seeds, less attention has been paid to this plant species by seed science researchers. Hence, this study was designed and performed to investigate the effects of different treatments on Galbanum seed dormancy-breaking.
Materials and Methods: In this study, two Galbanum seed populations were collected from Boyer-Ahmad pastures (Iran, Kohgiluyeh and Boyer-Ahmad Province) and Pakan Bazr Isfahan Company (Iran). Seeds were exposed to a cold stratification test for 120 days and germinability and dormancy breaking responses were studied. Also, seed drying effects on germinability after exposure to stratification treatment were investigated to evaluate the practical aspects of this treatment. The impacts of different hormonal pre-treatments including gibberellic acid (GA3) and cytokines (Kinetin, 6-Benzylaminopurine) at different concentrations on germination and dormancy behaviors were investigated so as to assess the effect of these treatments on seed dormancy breaking.
Results: Results showed that germination/dormancy behaviors of Galbanum populations were different and also the effect of different treatments on seeds germination and dormancy varied. An increase in germination was observed when stratification was applied for up to 60 days on two seed populations, thereafter germination decreased by prolonged stratification. Also, it was revealed that desiccation of the treated seeds after stratification reduced seed germination. Examining the cause of this issue revealed that the most important reason for the decrease in seed germination in long periods of stratification was the germination of seeds in the stratification bed and in other words, the difference in seed dormancy levels in different stratification periods. Seed germination behavior and dormancy breaking were improved significantly in both Galbanum populations when the different concentration of gibberellic acid and cytokinins was used.
Conclusion: The results of this study indicated the existence of different levels of seed dormancy in different Galbanum populations. It was also found that cold stratification and hormonal pre-treatment of seeds can be significantly effective in seed dormancy breaking. Eventually, it was concluded that Galbanum seed dormancy was classified as semi-deep and deep morphological dormancy due to the effects of different treatments.

Abstract

Extended Abstract:
 Introduction: Production of high-quality seeds to stabilize crop yield is an important challenge for breeders. One of the most important answers to this challenge is to clarify the molecular mechanisms associated with seed vigor characteristics. Functional proteins of Cupin superfamily are among the molecules in signaling pathway. Previous research has shown that in maize, a storage protein similar to the functional Cupin superfamily protein called ZmGLP is effective in seed germination. However, in the previous experiments, suitable indicators were not used to assess seed vigor and its relationship with field establishment. So, it is needed to study the performance of ZmGLP in predicting field emergence to complete the previous research.
 Materials and Methods: An experiment was performed on 14 samples of commercial inbred maize lines. In this experiment, in addition to the laboratory evaluation of seed germination, field indices of physiological seed quality including the percentage of seedling emergence in the field, time to 50% seedling emergence, time to 90% seedling emergence, seedling dry weight, seedling height and coefficient of variation of seedling height was also assessed. In the polymerase chain reaction, two pairs of primers (CF / CR primers and IDF / IDR primers) were used to identify the DNA sequence of the Cupin.
Results: The results show that the seeds were different in terms of physiological quality. The lowest percentage of germination in laboratory was related to K1264/1, while the lowest physiological quality of seeds in field indices was observed in K1263/17. The molecular test confirmed the presence of the desired allele at the InDel9 site of vigor-related genes in the three samples of B73, K1264/1, and K1264/5-1, but no amplification band of the InDel9 site was observed in all K1263/17 seed samples. Due to the fact that line K1264/1, which had the lowest germination percentage in the laboratory, had an amplification band at this related site to vigor, it is not enough to rely on the results of the laboratory germination test to investigate the relationship between this gene and seed vigor. The field emergence test and seed vigor test that have a good prediction of field emergence must be used in these studies.
Conclusions: According to the results of this experiment, molecular tests with functional markers based on Indel9 can be used to accelerate the evaluation of vigor, especially when the breeder is breeding a new line or hybrid. It is a useful, rapid, and effective molecular method to predict seed emergence in the field and screen the lines to ensure the genetic strength of the germination of the lines, especially in the temperate germplasms of corn. Finally, it is necessary to determine the threshold of low vigor during seed quality investigation in different cultivars, and relationship between the presence or absence InDel9 site should be considered in future research.

1- The feasibility of using molecular markers to determine the seed vigor of corn lines in the field was studied and optimized for the first time.
2- The results of physiological quality assessment of seeds in the field for the studies related to the relationship between molecular markers and seed vigor were exploited for the first time.
3- The Indel9 site and molecular markers related to seed vigor in the field were introduced.
