مقدمه: حبوبات، مهم­ترین منبع تأمین پروتئین گیاهی بوده و ماش سبز با تولید دانه­هایی حاوی درصد بالای پروتئین، از ارزش غذایی زیادی برای انسان برخوردار است. عمده مشکل شوری در جوانه‌زنی بذر گیاهان عالی در اثر مقادیر بیش‌ازحد کلرید سدیم، ایجاد فشار اسمزی، اختلال در جذب و انتقال یون‌های غذایی و اثرات مستقیم سمیت یونی روی غشـا و سیستم‌های آنزیمـی می‌باشد که در کل موجب کاهش جوانه‌زنی بذرها می‌گردند. استفاده خارجی از متیل جاسمونات می‌تواند اثرات ناشی از تنش­های مختلف از جمله شوری و خشکی را از طریق افزایش فعالیت آنتی‌اکسیدانی بذر تعدیل نماید. لذا هدف از این تحقیق ارزیابی اثر متیل جاسمونات و تیمار شوری بر ویژگی‌های جوانه‌زنی و فعالیت آنزیمی ماش سبز است.
مواد و روش‌ها: این آزمایش به‌صورت فاکتوریل بر پایه طرح کاملاً تصادفی با سه تکرار در آزمایشگاه گروه زراعت دانشگاه تربیت مدرس انجام شد. تیمارهای آزمایش شامل چهار پیش­ تیمار متیل جاسمونات، صفر، 50، 100 و 150 میلی مولار و چهار تیمار شوری، صفر، 2، 4 و 6 دسی­زیمنس بر متر شوری از منبع کلراید سدیم بود. پتری­ها در ژرمیناتور در دمای 25 درجه سلسیوس و در تاریکی کامل به مدت 14 روز قرار داده شدند. در این آزمایش صفات سرعت و درصد جوانه‌زنی، زمان رسیدن به 50 درصد جوانه‌زنی، آلفا و بتا آمیلاز، کاتالاز و پراکسیداز اندازه‌گیری شد.
یافته‌ها: نتایج آزمایش نشان داد که کم‌ترین شیب سرعت و درصد جوانه­زنی نهایی، در تیمارهای 50 و 100 میلی مولار محلول متیل جاسمونات حاصل شد. از نظر زمان لازم برای 50 درصد جوانه‌زنی، افزایش 7/4 روز به ازای افزایش هر یک دسی­زیمنس بر متر تنش شوری و کم‌ترین زمان آن در غلظت 79 میلی مولار محلول متیل جاسمونات برآورد شد. از نظر فعالیت آنزیم­های جوانه ­زنی، کاهش 031/0 میکرومول بر میلی‌لیتر بر دقیقه به ازای افزایش هر یک دسی­زیمنس بر متر تنش شوری و بیشترین مقدار آلفا آمیلاز 6/72 میکرومول بر میلی‌لیتر بر دقیقه در غلظت 73 میلی مولار محلول متیل جاسمونات تخمین زده شد. همچنین، کم‌ترین فعالیت آنزیم بتا آمیلاز 79/0 میکرومول بر میلی‌لیتر بر دقیقه در غلظت 57/5 دسی‌زیمنس بر متر تنش شوری و بیشترین فعالیت آنزیم بتا آمیلاز 3/1 میکرومول بر میلی‌لیتر بر دقیقه در غلظت‌ 87 میلی مولار محلول متیل جاسمونات برآورد شد. بیشترین فعالیت آنزیم کاتالاز (7/25 بر میلی‌گرم پروتئین بر دقیقه) در 72/14 دسی‌زیمنس بر متر تنش شوری و کم‌ترین فعالیت آنزیم کاتالاز (8/8 تغییرات جذب بر میلی‌گرم پروتئین بر دقیقه) در 88 میلی‌مولار محلول متیل جاسمونات برآورد شد. بیشترین فعالیت آنزیم پرکسیداز (06/22 تغییرات جذب بر میلی‌گرم پروتئین بر دقیقه) را در غلظت 30/24 دسی‌زیمنس بر متر تنش شوری و کم‌ترین فعالیت آنزیم پرکسیداز (5/2 تغییرات جذب بر میلی‌گرم پروتئین بر دقیقه) را در غلظت 267 میلی مولار محلول متیل جاسمونات تخمین زده شد.
نتیجه‌گیری: به ­طور کلی، کاربرد پیش‌تیمار متیل جاسمونات، از طریق بهبود فعالیت آنزیم­های جوانه­ زنی و افزایش فعالیت آنزیم‌های تنش، موجب افزایش فعالیت آنزیم­های هیدرولیزکننده در آندوسپرم بذرهای جوانه‌زده شده، که این امر سبب کاهش مدت زمان جوانه‌زنی، افزایش سرعت جوانه‌زنی و کاهش تنش اکسیداتیو در شرایط تنش شوری می‌گردد.

جنبه ­های نوآوری:
1- سرعت و درصد جوانه­زنی و تغییرات مورفولوژیک بذر ماش سبز تحت تأثیر متیل جاسمونات بررسی گردید.
2- نقش آنزیم­های جوانه­زنی آلفا و بتا آمیلاز در تسریع تولید گیاهچه ماش سبز تحت شرایط شور تخمین زده شد.
3- فعالیت آنزیم‌های کاتالاز و پراکسیداز ناشی از متیل جاسمونات در بروز مقامت به تنش شوری برآورد شد.
