fa تأثیر تیمارهای مختلف هورمونی و سرمادهی بر برخی شاخص‌های بیوشیمیایی و جوانه‌زنی بذرهای سه گونه مریم‌گلی (Salvia officinalis, S .leriifolia, S. syriaca) Effect of different hormonal and cold treatments on some biochemical and germination indices in seeds of three species of Salvia (Salvia officinalis, S. leriifolia, S. syriaca) فیزیولوژی بذر Seed Physiology پژوهشي Research <div style="text-align: justify;">چکیده مبسوط مقدمه: مریم‌گلی گیاهی ارزشمند و دارویی از خانواده نعناعیان دارای مصارف متعدد دارویی و غذایی است. پژوهش حاضر به‌منظور بررسی اثرات تیمار‌های هورمونی و دوره‌های سرمادهی مرطوب روی شاخص‌های بیوشیمیایی و جوانه‌زنی بذرهای سه گونه S .syriaca, S .leriifolia, S. officinalis انجام شد. مواد و روش‌ها: این تحقیق به صورت فاکتوریل در قالب طرح پایه کاملاً تصادفی با سه تکرار در سال 1403 و بطور دو بار مجزا انجام شد. سطوح سرمادهی مرطوب (A1-A4) به عنوان عامل اول و تیمارهای هورمونی (B1-B8) به عنوان عامل دوم قرار گرفتند. سطوح سرمادهی مرطوب شامل عدم سرمادهی، 10، 20 و 30 روز سرمادهی (دمای 4 درجه سلسیوس و رطوبت نسبی 45 درصد) بود. تیمارهای هورمونی شامل اسید جیبرلیک (120 میلی گرم در لیتر)، بنزیل آدنین (4-10 مولار)، کینتین (4-10 مولار)، اسید جیبرلیک (120 میلی‌گرم در لیتر)+ بنزیل‌آدنین (4-10 مولار)، اسید جیبرلیک (120میلی‌گرم در لیتر)+کینتین (4-10 مولار)، بنزیل آدنین (4-10 مولار) + کینتین (4-10 مولار)، اسید جیبرلیک (120 میلی‌گرم در لیتر) + بنزیل آدنین (4-10 مولار) + کینتین (4-10 مولار) و شاهد (آب مقطر) بودند. صفات مورد اندازه‌گیری در این آزمایش شامل شاخص‌های بیوشیمیایی مقادیر آنزیم‌های آلفا آمیلاز، بتا-1 و 3-گلوکوناز و پراکسید هیدروژن و شاخص‌های جوانه‌زنی از جمله طول و وزن خشک گیاهچه، درصد جوانه‌زنی، بنیه بذر، میانگین مدت جوانه‌زنی و شاخص جوانه‌زنی بودند. یافته‌ها: سرمادهی مرطوب و تیمارهای هورمونی به طور معنی‌داری بر صفات مورد ارزیابی اثرگذار بودند. اسید جیبرلیک در بیشتر صفات مورد بررسی، نقش مفید و افزایشی داشت. بیشترین و کمترین مقادیر صفات مهم جوانه‌زنی از جمله طول گیاه‌چه (56/5-1/2 سانتی‌متر)، شاخص بنیه بذر (1/5-94/0)، وزن خشک گیاهچه (35/45-84/19 میلی‌گرم)، جوانه‌زنی (74/91-22/38 درصد)، یکنواختی جوانه‌زنی (03/21-57/3 روز) و صفات بیوشیمیایی از جمله مقادیر آلفا آمیلاز (94/12-6/2 میلی‌مول بر گرم وزن تر بذر)، بتا-1 و 3-گلوکوناز (79/10-83/2 میلی‌مول بر گرم وزن تر بذر) و پراکسید هیدروژن (69/0-21/0 میلی‌مول بر گرم وزن تر بذر) به ترتیب در تیمار تلفیقی سرمادهی مرطوب (20 و 30 روز) + تیمار هورمونی بنزیل آدنین + کینتین + اسید جیبرلیک و تیمار عدم سرمادهی + عدم تیمار هورمونی به وجود آمد که نسبت به شاهد، افزایشی بیش از دو برابر را نشان داد. درصد جوانه‌زنی و غالب صفات مورد بررسی در گونه S. officinalis به ترتیب بیشتر از گونه S. syriaca و S. leriifolia بود. نتیجه‌گیری: مقادیر شاخص‌های بیوشیمیایی و جوانه‌زنی بذر گونه‌های مریم‌گلی متفاوت است. بر اساس برهمکنش سرمادهی و تیمارهای هورمونی، 20 روز سرمادهی مرطوب با کاربرد هر یک از تیمارهای بنزیل آدنین، کینتین و اسید جیبرلیک جهت جوانه‌زنی بذر مریم‌گلی و تولید بالاترین مقادیر شاخص‌های جوانه‌زنی و بیوشیمیایی، مناسب به نظر می‌رسد. جنبه‌های نوآوری: <ol> <li dir="RTL" style="margin-right:57px; text-align:justify">اثرات تیمارهای هورمونی و سرمادهی مرطوب روی شاخص‌های بیوشیمیایی و جوانه‌زنی بذر سه گونه مریم‌گلی بررسی شد.</li> <li dir="RTL" style="margin-right:57px; text-align:justify">20 روز سرمادهی مرطوب با کاربرد هر یک از تیمارهای بنزیل آدنین، کینتین و اسید جیبرلیک جهت جوانه‌زنی بذر مریم‌گلی پیشنهاد می‌گردد.</li> </ol> </div> <div style="text-align: justify;">Extended abstract Introduction: Salvia sp. from Lamiaceae family is one of the important medicinal and pasture plants that have many uses in various medicinal and food industries. This study was performed to evaluate the effects of different hormonal treatments on germination in three species of Salvia including S. officinalis, S. leriifolia, and S. syriaca under different wet stratification periods. Materials and Methods: This research was conducted in factorial layout experiment in a completely randomized design (CRD) with 3 replications and 2 separate repetitions in Herbal Research Institute, Shahrekord Branch, Islamic Azad University in 2024. Experimental treatments included wet chilling (0, 10, 20, 30 day in 4°C and RH=45%)  as first factor (A1-A4) and hormones inclusive (gibberellic acid (120ppm), benzyl adenine (10-4 M), kinetin (10-4 M), gibberellic acid (120 ppm) + benzyl adenine (10-4 M), gibberellic acid (120 ppm) + kinetin (10-4 M), benzyl adenine (10-4 M) + kinetin (10-4 M), gibberellic acid (120 ppm) + benzyl adenine (10-4 M)+ kinetin (10-4 M) and control) as second factor (B1-B8). Biochemical indices such as hydrogen peroxide, α-amylase and beta-1,3glucanase and germination indices such as dry weight of seedlings, length of seedlings, germination percentage, mean of germination time, seed vigour, and germination uniformity were measured. Results: The use of wet chilling and hormonal treatments had significant effectiveness on germination characters. Gibberellic acid in the most of measured characters had beneficial and increasing effects. The most and lowest important measured germination characteristics and decreased time needed for germination. The highest and lowest amounts for germination characteristics such as seedling length (5.56-2.1 cm), seed vigor (5.1-0.94), seedling dry weight (45.35-19.84 mg), germination percentage (91.74-38.22), germination uniformity (21.03-3.57 day) and biochemical characters such as content of α-amylase (12.94-2.6 mM/g FW of seed), beta-1, 3 glucanase (10.79-2.83 mM/g FW of seed) and hydrogen peroxide (0.69-0.21 mM/g FW of seed) were recorded under wet chilling treatment for 20 day + gibberellic acid (120 ppm) + benzyl adenine (10-4 M) + kinetin (10-4 M) and treatment of without wet chilling + without hormonal treatment, respectively. Also, increasing wet chilling treatment duration, increased some evaluated characteristics, but most of them reached the highest values after 20 days wet chilling treatment. The amounts of evaluated characters in this treatment improved more than 100% in comparison to control seeds. It can be concluded that wet chilling (20 day) along with the use of gibberellic acid, benzyl adenine, and kinetin resulted in improved seed germination indices of Salvia spp. Results showed that in most of measured characteristics, the Salvia species have various amounts of biochemical and germination indices. Finally, the treatment of wet chilling (20 day) and use of gibberellic acid, benzyl adenine, and kinetin hormones, suggest to obtain of the best germination indices in seeds of Salvia spp. Germination percentage and most of the other measured characters in this research in S. officinalis were more than S. syriaca, and S. leriifolia, respectively. Conclusion: Results showed that the most of measured characteristics of Salvia species have various amounts of biochemical and germination indices.
