fa بررسی شرایط بهینه دما، نور، مدت و بستر کشت برای جوانه‌زنی بذر سه گونه سالیکورنیا Study on optimal temperature, light, duration, and substrate conditions for three Salicornia species seed germination سایر موضوعات General پژوهشي Research <div style="text-align: justify;">چکیده مبسوط مقدمه: سالیکورنیا گیاهی شورپسند است که کشت آن برای اصلاح اراضی شور و تولید علوفه از آن از اهمیت برخوردار است. جوانه زنی بذر به دلیل نقش کلیدی آن در استقرار جمعیت گیاهی همیشه مورد توجه بومشناسان گیاهی بوده است. همچنین به علت اهمیت این فرآیند در گواهی بذر، این پدیده مورد توجه متخصصین کنترل و گواهی بذر است. دما و دسترسی به رطوبت کافی و نیز وجود نور در گونه های حساس به نور برای جوانهزنی بذر از مهمترین عوامل طبیعی برای رخداد جوانه زنی بذر محسوب می گردند. همچنین مدت زمان لازم برای جوانه زنی بذر و رشد اولیه کافی گیاهچه برای تعیین قابلیت جوانه زنی بالقوه بذر مهم است. از اینرو تعیین دما، نیاز یا عدم نیاز به نور و نیز مدت زمان لازم برای جوانه زنی و  نیز بستر مناسب برای کاشت بذرها در فرآیند آزمون های آزمایشگاهی گواهی بذر از اهمیت به سزائی برخوردار هستند. این آزمایش به منظور بررسی شرایط بهینه دما، نور، مدت و بستر کشت برای جوانه‌زنی بذر سه گونه سالیکورنیا انجام شد. مواد و روش‌ها: بررسی اولیه برای تعیین نیاز جوانه‌زنی بذر گیاه سالیکورنیا پرسیکا، پرسپولیتانا و بیگلوی به نور، مدت زمان و بسترهای کشت مناسب برای آزمون جوانه‌زنی استاندارد، یک آزمایش مقدماتی  انجام شد. با توجه به عدم مشاهده تفاوت درصد گیاهچه‌های ظاهر شده در معرض نور و تاریکی (بذرهای گونه‌های مورد بررسی سالیکورنیا تحت شرایط روشنایی و بدون نور جوانه زدند) و نیز رسیدن به حداکثر جوانه‌زنی بذرها در مدت 7 و 12 روز در بسترهای کشت بین کاغذ جوانه‌زنی (BP) و روی کاغذ جوانه‌زنی (TP)، در دماهای 20 و 25 به‌صورت ثابت و دمای 25-20 درجه سلسیوس متناوب (16- 8 ساعت / روز- شب)، آزمایش اصلی تحت این شرایط به اجرا درآمد. یافته‌ها: نتایج نشان داد بذرهای گونه بیگلوی دارای بیشترین درصد گیاهچههای عادی در دمای 25 درجه سلسیوس بهصورت ثابت در مدت 7 روز در بستر کشت روی کاغذ جوانهزنی (TP) بودند. همچنین بذرهای گونه پرسیکا دارای بالاترین درصد گیاهچه‌های عادی در دمای 20-25 درجه سلسیوس متناوب بهمدت 7 روز در بستر کشت روی کاغذ جوانه زنی (TP) بودند. بذرهای گونه پرسپولیتانا در دمای 20 درجه سلسیوس به صورت ثابت به مدت 7 روز روی بستر کشت کاغذ جوانهزنی (TP) از بیشترین درصد گیاهچه های عادی برخوردار بودند. در دماهای جوانه زنی و بسترهای کشت مورد بررسی در هر دو مدت جوانه زنی به ترتیب بذرهای گونه پرسیکا، بیگلوی و پرسپولیتانا درصد گیاهچه های عادی بالاتری داشتند.  نتیجه‌گیری: نتایج این تحقیق نشان داد، بذرهای گونههای مورد بررسی سالیکورنیا برای جوانهزنی به نور نیاز نداشتند. همچنین گونه های بررسیشده سالیکورنیا در این تحقیق از لحاظ دما، مدت و بستر کشت بهینه برای آزمون جوانه زنی استاندارد با یکدیگر تفاوت معنیدار داشتند. به طوریکه دمای بهینه برای جوانه زنی بذرهای گونه پرسیکا دمای متناوب و دمای بهینه برای جوانه زنی بذرهای گونه های بیگلوی و پرسپولیتانا دمای ثابت بود. دمای ثابت برای جوانهزنی بذرهای گونه بیگلوی بیش از دمای ثابت برای جوانه زنی بذرهای گونه پرسپولیتانا بود. همچنین بستر کشت روی کاغذ جوانه زنی (TP) برای آزمون جوانه زنی استاندارد هر سه گونه مناسب بود. جنبه‌های نوآوری: <ol> <li class="CxSpMiddle" dir="RTL" style="margin-right:47px; text-align:justify">جوانه زنی بذرهای گونههای مورد بررسی سالیکورنیا نیازمند به نور نبود.</li> <li class="CxSpMiddle" dir="RTL" style="margin-right:47px; text-align:justify">پاسخ جوانه زنی بذرهای گونههای مورد بررسی سالیکورنیا به دما و مدت زمان جوانه‌زنی مطلوب متفاوت بود.</li> <li class="CxSpMiddle" dir="RTL" style="margin-right:47px; text-align:justify">گونه های مورد بررسی سالیکورنیا ازلحاظ بستر کشت مناسب برای جوانه زنی بذر تفاوت معنیداری نداشتند.</li> </ol> </div> <div style="text-align: justify;">Extended abstract Introduction: Salicornia is a halophyte plant which cultivation is important for reclamation of saline soils and producing fodder. Seed germination has always been of interest to plant ecologists due to its key role in plant population establishment. Also, due to the importance of this process in seed certification, this phenomenon is of interest to control and seed certification experts. Temperature, access to sufficient humidity, and the presence of light in light-sensitive species for seed germination are considered to be the most important natural factors for seed germination. Additionally, the time required for germination and sufficient early seedling growth are important to determine the potential seed germination. Therefore, determining the temperature, the need or lack of light, as well as the time required for germination and the suitable substrate for planting seeds, are of great importance in the process of seed certification laboratory tests. This experiment was conducted to investigate the optimal conditions of temperature, light, duration, and substrate for seed germination of three Salicornia species. Materials and Methods: A preliminary study was conducted to determine the light requirements, duration, and suitable substrate for the standard germination test of Salicornia persica, S. persepolitana, and S. bigelovi, seeds. Since no difference was observed in the percentage of seedlings emerging in light and darkness (seeds of the studied Salicornia species germinated under light and dark conditions) and maximum seed germination was achieved within 7 and 12 days in the substrate between germination paper (BP) and top of paper (TP), at constant temperatures of 20°C and 25 °C and alternating temperatures of 20-25 °C (8-16 hours/day-night), the main experiment was carried out under these conditions. Results: The results showed that the seeds of S. bigelovi species had the highest percentage of normal seedlings at 25 °C constant temperature for 7 days in the top-of-paper (TP) substrate. Also, the seeds of S. persica had the highest percentage of normal seedlings at 20-25 °C alternating temperature for 7 days in the top-of-paper (TP) substrate. S. persepolitana seeds at 25 °C constant temperature for 7 days on the top of paper (TP) substrate had the highest percentage of normal seedlings. S. persica, S. bigelovi, and S. persepolitana seeds had a higher percentage of normal seedlings in both germination durations and temperatures, respectively. Conclusions: The results of the research showed that the seeds of the studied Salicornia species did not require light for germination. Also, the studied Salicornia species in the research had significant differences in terms of temperature, duration, and optimal substrate for the standard germination test. So that the optimal temperature for germination of S. persica seeds was alternative temperature and the optimal temperature for germination of S. bigelovii and S. perspolitana seeds were constant temperature. The constant temperature for germination of S. bigelovii species seeds was higher than the constant temperature for germination of S. perspolitana seeds. Also, the top of paper (TP) substrate was suitable for the standard germination test of all three species.
