fa جوانه‌زنی و ضرایب حیات بذر باقلا (Vicia faba) در پاسخ به شرایط مختلف انبارداری Germination and viability coefficients of Faba Bean (Vicia faba) seed under different storage conditions فیزیولوژی بذر Seed Physiology پژوهشي Research <div style="text-align: justify;">چکیده مبسوط مقدمه: حبوبات از منابع مهم غذایی سرشار از پروتئین و یکی از اجزاء اصلی در تناوب زراعی است که در سال‌های اخیر، به‌عنوان یکی از گزینه‌های عمده تحقیقات گیاهی به‌شمار می‌آید. قابلیت انبارداری بذر یکی از صفات مهم در اصلاح حبوبات می‌باشد. دما، محتوی رطوبت بذر و طول دوره انبارداری از مهم‌ترین عوامل مؤثر بر کیفیت بذر در مدت انبارداری هستند. شرایط نامساعد انبارداری منجر به زوال و کاهش کیفیت بذرها طی نگهداری می‌شود که به‌شدت تحت تأثیر شرایط محیطی انبارداری قرار می‌گیرد. مواد و روش‌ها: این پژوهش در آزمایشگاه فناوری بذر دانشکده کشاورزی دانشگاه یاسوج در سال 1393 به صورت فاکتوریل سه عاملی در قالب طرح کاملاً تصادفی، با پنج تکرار 20 بذری اجرا شد. بذرها با محتوی رطوبت در 5 سطح 6، 10، 14، 18 و 22 درصد و دما در 4 سطح 15، 25، 35 و 45 درجه سلسیوس به‌مدت 9 ماه (صفر، 30، 60، 90، 120، 150، 180، 210، 240 و 270 روز) نگهداری شدند. پس از هر بار نمونه‌گیری در پایان هر ماه انبارداری، آزمون جوانه‌زنی استاندارد درون ژرمیناتور با دمای 25 درجه سلسیوس به‌مدت 10 روز و به روش کاغذ آکاردئونی انجام شد. همچنین آزمون هدایت الکتریکی مواد نشت یافته از بذرهای خیسانده شده به‌مدت 24 ساعت در دمای 20 درجه سلسیوس درون ژرمیناتور نیز در چهار تکرار انجام گرفت. برخی صفات جوانه‌زنی و هدایت الکتریکی مواد نشت یافته از بذر طبق روش‌های استاندارد اندازه‌گیری شدند. یافته‌ها: نتایج نشان داد، اثر سه‌گانه دمای انبارداری، محتوی رطوبت بذر و طول دوره انبارداری بر شاخص‌های جوانه‌زنی و هدایت الکتریکی بذر باقلا در سطح احتمال یک درصد معنی‌دار بود. روند جوانه‌زنی طی انبارداری در دمای 15 درجه سلسیوس و محتوی رطوبت بذر 6 درصد پس از 270 روز انبارداری از 94 درصد به 81 درصد کاهش یافت. به‌طوری‌که در شرایط رطوبتی مشابه با افزایش دما از 15 به 45 درجه سلسیوس پس از 270 روز انبارداری، جوانه‌زنی به 35 درصد کاهش یافت. با گذشت زمان انبارداری، هدایت الکتریکی مواد نشت یافته از بذر افزایش یافت و در دماهای بالا این افزایش بیشتر بود. ضریب‌های حیات بذر باقلا نیز پس از 9 ماه انبارداری با استفاده از معادله قابلیت حیات بذر محاسبه شد که KE،CW ، CH  و CQ به‌ترتیب 39697/5-، 13041/2، 03201/0 و 000017/0 محاسبه گردید. نتیجه‌گیری: نتایج کلی آزمایش نشان داد که با افزایش دما و رطوبت بذر در مدت انبارداری هدایت الکتریکی مواد نشت یافته از بذر افزایش داشت که این مسئله منجر به کاهش درصد زنده‌مانی بذر شد. دمای 15 درجه سلسیوس و محتوای رطوبت بذر 6 درصد نسبت به تمامی دما و رطوبت‌ها شرایط مناسب‌تری را برای زنده‌مانی بذرها در طول دوره 9 ماهه انبارداری داشتند به‌طوری‌که دارای کمترین هدایت الکتریکی مواد نشت یافته از بذر و در نهایت سرعت زوال بودند.  جنبه‌های نوآوری:</div> <ol> <li dir="RTL" style="text-align: justify;">با گذشت زمان انبارداری، هدایت الکتریکی مواد نشت یافته از بذر افزایش یافت.</li> <li dir="RTL" style="text-align: justify;">هدایت الکتریکی مواد نشت یافته از بذر با محتوی رطوبت 22 درصدی و دمای انبارداری 45 درجه سلسیوس افزایش داشت.</li> <li dir="RTL" style="text-align: justify;">ضریب‌های حیات بذر باقلا جهت ارزیابی قابلیت حیات بذر در شرایط انبارداری کنترل شده محاسبه گردید.</li> </ol> Extended Abstract Introduction: Pulses are among the best sources of plant protein and important components of crop rotation, which in recent years, have been considered one of the major options for plant research. Seed storage is one of the important traits in legume breeding. Storage temperature, seed moisture content, and storage duration are the most important factors affecting seed quality during storage. Inappropriate storage conditions lead to deterioration and reduction of seed quality during storage, which is severely affected by the environmental conditions of storage. Materials and Methods: This research was conducted at the Seed Technology Laboratory, Faculty of Agriculture, Yasouj University in 2014 as a three-way factorial based on the completely randomized design with 5 replications of 20 seeds. Seeds with moisture content at 5 levels (6, 10, 14, 18, and 22%) and storage temperature at 4 levels (15, 25, 35 and 45 °C) were stored for 9 months (0, 30, 60, 90, 120, 150, 180, 210, 240 and 270 days). After sampling at the end of each month, a standard seed germination test was done using the pleated paper method in a germinator at 25 °C for 10 days. Also, an electrical conductivity test of the electrolytes leaked from the seeds incubated for 24h in water at 20 ˚C was done with 4 replicates. Some germination attributes and electrical conductivity of the electrolytes leaked from the seeds were measured according to standard methods. Results: According to the results, interaction effects of storage temperature, seed moisture content, and storage duration on germination indices and electrical conductivity of bean seeds were significant (P<0.1). The germination trend during storage at 15 °C and seed moisture content of 6% decreased from 94% to 81% after 270 days of storage, so that germination decreased to 35% under similar moisture content after 270 days of storage as temperature increased from 15 to 45 °C. As the storage time passed, electrical conductivity increased and this increase was more pronounced at higher temperatures. Viability constants were calculated 9 months after storage using the seed viability equation, in which KE, CH, CW, and CQ were calculated -5.39697, 0.03201, 2.13041, and 0.000017, respectively. Conclusions: The results showed that the electrical conductivity of the leaked material increased with increasing storage temperature and seed moisture content, which led to lower viability of seeds. At 15 °C and 6% seed moisture content provided better conditions for seed survival during the 9-month storage time compared with all other temperatures and moistures and had the lowest rate of deterioration. The results showed that with increasing seed temperature and moisture so that they had to lowest electrical conductivity of the leaked material from seeds and deterioration rate.
